Yea, rebooting or turning off the computer will pretty much clear everything stored in RAM. (Although I've heard the FBI can recover it if needed) If you're talking about "defragmenting", testing, or freeing up extra memory, you might try some of the tools on this page:

The only one I've actually had personal experience with though is MemTest86, although I have seen a RAM "clearing" program similiar to others listed run on a windows 98 machine that worked.
If you need more free RAM though, the solutions is to just buy more and install it (or have it professionally installed, but it isn't hard)
